This Romulus lair entrance is found at the base of the massive wall that surrounds the city, at the south end of the map. Most of the initial platforming is all pretty obvious, moving you forward along a linear path. But soon things get less linear, as the path branches in multiple directions. Fortunately, there is a way to get through the mazes quickly. Always follow the torches.
Torches on the walls will always be near to guide you down the correct path. As long as you follow the torches, you'll never get lost. You soon come to a large well that you need to climb down carefully. Jump from the wooden plank at the top to the opposite side of the well and tap the grab button to drop down. Tap the grab button while falling to grab a ledge. When you get near the bottom, slowly climb down to a landing from which you can turn around and jump to the next area.
Continue following torches to get through the mazes. When you reach a massive hall with a few followers of Romulus that scamper away, climb up the wooden structure on your left. You need to follow a series of jumps and hand holds around the eastern perimeter of the hall before turning west toward a giant cross. Climb the cross and jump west off the top of it to continue.
Finally, you'll enter a circular room with a group of enemies. Fight off the crew on the bottom row and then look for a ladder that'll bring you to the second level with more enemies. You need to hop from the outer walkway onto the center column of platforms, jumping back and forth between them to reach the top. You'll encounter enemies along the way but if you're trying to beat this stage quickly you can do your best to ignore them and continue to the end of the lair. I found this area particularly annoying, as you can fall if the enemies throw stones at you.

Be careful not to fall from up high. To get full sync in this mission, you can't lose more than one block of health. There are no enemies to fight, but falling from significant distance will basically end your ability to get full sync. When you reach an area with a statue of a woman with her hands out, climb up onto the statue and jump left onto a hanging lantern. As Ezio is flung off the lantern, you need to manually tap the grab button to latch onto some wood surrounding a column.
From there you can shimmy around and continue. You'll swing down to a lower level before climbing back to the upper level and heading west. In a large room with a floor that gives out as you enter, hug the left south wall and use a thin, rickety wooden structure attached to the wall to grab a ledge ahead.
Soon you'll enter the final area, a massive Cathedral hall. Run to the north end and interact with a glowing gate. You need to equip a ranged weapon to hit a hanging sand bag left of the gate. Finding your way to the treasure at the end of each lair is reasonably easy, but can you do it like Ezio would have and achieve full synchronization? For the timed lair runs, a good tip to remember is to position the camera in the direction you intend to leap before making each jump.
The trick to making this speed run is to simply remember where to start a few of the key platforming sections. Then of course, make sure not to fall. Keeping your health up should be no problem here.
